Putrescine and spermidine biosynthesis in the development of normal and anucleolate mutants of Xenopus laevis.

چکیده
Ornithine decarboxylase (EC 4.1.1.17), the enzyme that catalyzes the synthesis of putrescine from ornithine, increases dramatically in developing Xenopus embryos. Between the 2-cell stage and early blastula stage, activity increases 10-fold, and in swimming tadpoles, the enzyme activity is 100-fold higher than that present in either unfertilized eggs or 2-cell embryos. S-adenosyl-L-methionine decarboxylase, an enzyme that catalyzes spermidine synthesis from putrescine and S-adenosyl-L-methionine, increases 40-fold in activity during the development of Xenopus, but does not increase in activity prior to gastrulation. Concomitant with these enzyme changes, putrescine and spermidine concentrations are elevated during the development of Xenopus embryos. Maximal accumulations are present in the swimming tadpole and correspond to maximal enzyme activities. Anucleolate-mutant embryos of Xenopus, which do not synthesize new ribosomes, have no detectable S-adenosyl-L-methionine decarboxylase activity and do not accumulate spermidine after gastrulation. Ornithine decarboxylase activity is depressed in these mutants and putrescine accumulation is decreased also. The activity of some dehydrogenases that increase in Xenopus embryos after gastrulation show normal increases in the anucleolate mutants. Thus, the synthesis of putrescine and spermidine in embryos correlates with the onset of ribosomal-RNA synthesis and the formation of a viable nucleolus in the embryonic cell.

The development of anucleolate embryos of Xenopus laevis.
A MUTATION of Xenopus has been found which reduces the maximum number of nucleoli per diploid nucleus from 2 in the wild-type to 1 in heterozygotes. Homozygous mutants possess no true nucleoli, hence being termed anucleolate. They do, however, possess pyronine-staining intranuclear organelles that are smaller and more numerous than typical nucleoli. The mutation can be considered as a recessive...
متن کاملExpression of hsp90 Alpha and hsp90 Beta during Xenopus laevis Embryonic Development
Background: Members of the eukaryotic Hsp90 family function as important molecular chaperones in the assembly, folding and activation of cellular signaling in development. Two hsp90 genes, hsp90 alpha and hsp90 beta, have been identified in fish and homeothermic vertebrates but not in poikilothermic vertebrates. In the present study, the expression of hsp90 alpha and hsp90 beta genes in Xenopus...
متن کاملControl of 5s Rna Synthesis during Early Development of Anucleolate and Partial Nucleolate Mutants of Xenopus Laevis
Ribosomes of all eukaryotes contain a single molecule of 5S, 18S, and 28S RNA. In the frog Xenopus laevis the genes which code for 18S and 28S RNA are located in the nucleolar organizer, but these genes are not linked to the 5S RNA genes. Therefore the synthesis of the three ribosomal RNAs provides a model system for studying interchromosomal aspects of gene regulation. In order to determine if...

Abstract The S362A mutation block ROMK2 (Kir1.1b) endocytosis in Xenopus laevis oocyte membrane . Saeed Hajihashemi1 , 1-Assistant professor, PhD in Physiology, Department of Physiology, School of Medical science, Arak University of Medical Sciences. Introduction: ROMK channel is localized on the apical membrane of the nephron. Recent studies suggest that endocytosis of ROMK chan...
متن کاملIntegrin Linked Kinase (X-ILK) Function during Embryonic Development and within Adult Tissues of Xenopus laevis
Integrin linked kinase (ILK) is a serine/threonine protein kinase implicated in the phosphatidylinositol 3’kinase (PI3’K) pathway. Integrin linked kinase has been investigated in different organisms such as mammalian systems (human, mice, rat), insects (Drosophila) and nematodes (Cenorhabditis elegans), however to date little data regarding ILK research on amphibians has been reported. In...
